Specificity

Clone 2C6 is a ZooMAb® rabbit recombinant monoclonal antibody that detects GRK2. It targets an epitope within 23 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal half of human GRK2.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GRK2 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected GRK2 in lysates from THP-1 cells, mouse brain tissue, and rat spleen tissue, human spleen tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected GRK2 in human spleen tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with GRK2 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Beta-adrenergic receptor kinase 1 (UniProt: P25098; EC:2.7.11.15 also known as Beta-ARK-1, G-protein coupled receptor kinase 2) is encoded by the GRK2 (also known as ADRBK1, BARK, BARK1) gene (Gene ID: 156) in human. GRK2 is a member of the G protein-coupled receptor kinase (GRK) family. It consists of an N-terminal domain that interacts with G protein-coupled receptors (GPCRs), a central catalytic domain responsible for its kinase activity, and a C-terminal domain that mediates interactions with other proteins and lipids. GRK2 plays a crucial role in the regulation of GPCR signaling. Upon activation of GPCRs, GRK2 phosphorylates specific serine and threonine residues on the receptor, leading to the recruitment of arrestin proteins. This process desensitizes the receptor, preventing further G protein activation and initiating receptor internalization and downregulation. GRK2 thus modulates the intensity and duration of GPCR signaling, impacting various physiological processes. GRK2 is implicated in several diseases. In cardiovascular diseases, such as heart failure, elevated levels of GRK2 are associated with impaired β-adrenergic receptor signaling, contributing to decreased cardiac function. Additionally, GRK2 has been implicated in metabolic disorders, inflammatory diseases, and certain cancers, where its dysregulation can alter cellular signaling pathways, affecting disease progression and response to treatment. Modulating GRK2 activity is being explored as a therapeutic strategy in these conditions. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
