General description

Bromodomain-containing protein 2 (UniProt: P25440; also known as O27.1.1) is encoded by the BRD2 (also known as KIAA9001, RING3) gene (Gene ID: 6046) in human. Bromodomain-containing protein 2 (BRD2) is a member of the bromodomain and extraterminal (BET) protein family, functioning as a pivotal epigenetic regulator by recognizing acetylated lysine residues on histones, thereby modulating chromatin structure and gene expression. BRD2 contains two tandem N-terminal bromodomains that facilitate its binding to acetylated histones, particularly histone H4. The C-terminal conserved extra-terminal domain is crucial for protein-protein interactions. BRD2 is ubiquitously expressed across various tissues. BRD2 is implicated in numerous cellular processes, including cell cycle progression, apoptosis, and immune response modulation, and regulation of alternative splicing of several hundred genes. Its dysregulation contributes to tumorigenesis by influencing oncogene expression and maintaining genomic stability. BRD2 also involved in neurological disorders, inflammatory diseases and BRD2 mutants have been associated with increased susceptibility to juvenile myoclonic epilepsy. Specificity

Clone 1M5 is a ZooMAb® rabbit recombinant monoclonal antibody that detects BRD2. It targets an epitope within 22 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal region of human BRD2.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected BRD2 in HEK293 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected BRD2 in lysate from mouse brain tissue and a 1:1,000 dilution from a representative lot detected BRD2 in lysate from rat brain tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:2,000 dilution from a representative lot detected BRD2 in human testis tissue sections.

Immunocytochemistry Analysis: A 1:100 from a representative lot detected BRD2 in HEK-293 cells.

Affinity Binding Assay: A representative lot of this antibody bound with BRD2 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
