General description

3-beta-hydroxysteroid dehydrogenase/Delta 5-->4-isomerase type 1 (UniProt: P14060; also known as 3 beta-hydroxysteroid dehydrogenase/Delta 5-->4-isomerase type I (3-beta-HSD I), 3-beta-hydroxy-5-ene steroid dehydrogenase, 3-beta-hydroxy-Delta(5)-steroid dehydrogenase (EC:1.1.1.145), 3-beta-hydroxysteroid 3-dehydrogenase (EC:1.1.1.270), Delta-5-3-ketosteroid isomerase) is encoded by the HSD3B1 (also known as 3BH, HSDB3A) gene (Gene ID: 3283) in human. HSD3B1 is an essential enzyme involved in steroid biosynthesis, catalyzing the conversion of 5-steroid precursors into their 4-keto forms, which are critical intermediates in the production of progesterone, androgens, and estrogens. HSD3B1 is a member of the short-chain dehydrogenase/reductase family, featuring a conserved NAD+-binding domain required for its enzymatic activity. Expression of HSD3B1 is tissue-specific, predominantly localized in steroidogenic tissues such as the adrenal glands, gonads, and placenta. It is found in the smooth endoplasmic reticulum, where steroid biosynthesis occurs. HSD3B1 interacts with other enzymes in steroidogenic pathways, such as CYP17A1, facilitating androgen and estrogen synthesis. Genetic polymorphisms, such as the HSD3B1 (1245C) variant, have been associated with altered enzymatic activity, leading to increased androgen synthesis and resistance to androgen-deprivation therapy in prostate cancer. Dysregulation of HSD3B1 expression or activity is implicated in diseases such as metastatic castration-resistant prostate cancer, congenital adrenal hyperplasia, and hormone-related disorders. KLH-conjugated linear peptide corresponding to 17 amino acids from the internal region of human HSD3B1.

Application

Quality Control Testing

Evaluated by Western Blotting in Caco-2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected HSD3B1 in Caco-2 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HSD3B1 in human placenta tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected HSD3B1 in human placenta tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with HSD3B1 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 2M19 is a ZooMAb® rabbit recombinant monoclonal antibody that detects HSD3B1. It targets an epitope within 17 amino acids from the internal region.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
