General description

Reticulon-4 receptor (UniProt: Q9BZR6; also known as Nogo receptor (NgR), Nogo-66 receptor) is encoded by the RTN4R (also known as NOGOR, UNQ330/PRO526) gene (Gene ID: 65078) in human. The Nogo receptor (NgR) is a glycophosphatidylinositol (GPI)-anchored cell-surface protein that plays a significant role in axonal growth inhibition and neural plasticity. It is characterized by eight leucine-rich repeats (LRRs) flanked by N-terminal and C-terminal disulfide-containing cap domains, which contribute to its stability and ligand binding specificity. These LRRs are crucial for interactions with myelin-associated inhibitors such as Nogo-A, oligodendrocyte myelin glycoprotein (OMgp), and myelin-associated glycoprotein (MAG), which mediate the suppression of axon regeneration in the central nervous system (CNS). The receptor also contains a C-terminal stalk region and is anchored to the cell membrane via a GPI linkage, which positions it optimally for extracellular ligand interactions and signaling. The Nogo receptor is critical regulator of axonal growth and neural plasticity, primarily studied in the context of neuroregeneration and central nervous system (CNS) development. The Nogo receptor interacts with ligands such as Nogo-A, a myelin-associated inhibitor, to inhibit axon sprouting and regeneration, restricting neuroaxonal plasticity in adult mammals. This receptor is predominantly expressed in neurons and plays a key role in limiting recovery after CNS injuries, such as spinal cord injury (SCI). Recent studies have highlighted its role in modulating neuroinflammation, mitochondrial function during conditions like multiple sclerosis, and its impact on amyloid-β (Aβ) production in Alzheimer s disease pathogenesis. Additionally, NgR signaling has been implicated in regulating neurite outgrowth, precursor migration, and branching during nervous system development. The Nogo receptor also plays non-neuronal roles, such as regulating macrophage polarization and recruitment during inflammatory responses, further broadening its significance in neurodegenerative diseases and systemic inflammation. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Application

Quality Control Testing

Evaluated by Western Blotting in Neuro-2a cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Nogo Receptor in Neuro-2a cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Nogo Receptor in A549 cell lysate.

Enzyme Immunoassay: A representative lot of this antibody detected Nogo Receptor protein in ELISA application.

Affinity Binding Assay: A representative lot of this antibody bound with Nogo Receptor peptide with at least one million-fold (1,000,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1E18 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Nogo Receptor. It targets an epitope within 20 amino acids from the C-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
