Specificity

Clone 4G22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Endoplasmin/GRP94. It targets an epitope within 18 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the C-terminal region of human Endoplasmin/GRP94.

Application

Quality Control Testing

Evaluated by Western Blotting in C2C12 cell lysates.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Endoplasmin/GRP94 in C2C12 cell lysates.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Endoplasmin/GRP94 in COS-7 and PANC-1 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound Endoplasmin/ GRP94 peptide with a KD of 2.1 x 10-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Endoplasmin/GRP94 in human thyroid tissue sections.

Flow Cytometry Analysis: 1 μg from a representative lot detected Endoplasmin/GRP94 in one million C2C12 cells.

Target description

Endoplasmin (UniProt: P14625; also known as 94 kDa glucose-regulated protein, GRP-94, Heat shock protein 90 kDa beta member 1, Tumor rejection antigen 1, gp96 homolog) is encoded by the GRP94 (also known as HSP90B1, TRA1) gene (Gene ID: 7184) in human. GRP94 is a disulfide-linked homodimeric calcium-binding protein that is confined to the lumen of endoplasmic reticulum (ER). It serves as a chaperone for secreted and membrane proteins. It is targeted to the ER by its 21 amino acid N-terminal signal sequence that is subsequently cleaved off upon entry to the lumen, exposing the DDEV terminus of the mature protein. It maintains its presence in the ER by using its C-terminal sequence KDEL that serves as an ER retrieval signal for the KDEL receptor. GRP94 shares many biochemical features with HSP90, in particular its domain structure and ATPase activity. However, its mode of action varies from HSP90 in the conformational changes induced by nucleotide binding, and in its interactions with co-chaperones, which are very different from known cytosolic co-chaperones. It is reported to be more selective than many of the ER chaperones. GRP94 may also participate in the unfolding of cytosolic proteins lacking the secretion signal sequence. Murine models lacking GRP94 are reported to undergo development arrest at gestational day 7, before gastrulation and formation of mesodermal tissues. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
