General description
Lymphatic vessel endothelial hyaluronic acid receptor 1 (UniProt: Q9Y5Y7; LYVE-1, Cell surface retention sequence-binding protein 1, CRSBP-1, Extracellular link domain-containing protein 1, Hyaluronic acid receptor) is encoded by the LYVE1 (also known as CRSBP1, HAR, XLKD1, UNQ230/PRO263) gene (Gene ID: 10894) in human. LYVE-1 is a disulfide-linked, homodimeric, single-pass type I membrane protein that undergoes ligand-dependent internalization and recycling at the cell surface. It serves as a ligand-specific transporter trafficking between intracellular organelles (TGN) and the plasma membrane. It is synthesized with a signal peptide (aa 19), which is subsequently cleaved off to produce the mature protein that contains a large extracellular domain (aa 20-238), a transmembrane Helical domain (aa 239-259), and a cytoplasmic tail (aa 260-322). It is mainly expressed in endothelial cells lining lymphatic vessels. It is expressed on both the luminal and basolateral surfaces of lymphatic endothelium. It is reported to mediate the uptake of intermediate and high molecular weight hyaluronan (HA) and serves as a low affinity receptor for HA-induced lymphoangiogenesis. It also controls the entry and/or migration of leukocytes in lymphatic vessels. The capacity of LYVE-1 to bind HA is based on a receptor distribution and organization of the glycosaminoglycans. Application
Quality Control Testing
Evaluated by Immunohistochemistry by human liver tissue sections.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LYVE1 in human liver tissue sections.
Tested Applications
Flow Cytometry Analysis: 1 µg from a representative lot detected LYVE1 in one million human umbilical vein endothelial cells (HUVEC).
Affinity Binding Assay: A representative lot of this antibody bound LYVE1 peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LYVE1 in MCF7 cells.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biochem/physiol Actions
Clone 2F9 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects LYVE1. It targets an epitope within 21 amino acids from C-terminal region of extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
