Specificity

Clone 1B9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Syntaxin 4. It targets an epitope within 19 amino acids from the C-terminal half.
Clone 3G10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects BLAST-2/CD23. It targets an epitope within 25 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal half of human Syntaxin 4.
KLH-conjugated linear peptide corresponding to 25 amino acids from the extracellular domain of human BLAST-2/CD23.

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in Human tonsil tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected BLAST-2/CD23 in Human tonsil tissue sections.

Tested Applications

Flow Cytometry Analysis: 1 µg from a representative lot detected BLAST-2/CD23 in one million Raji cells.

Affinity Binding Assay: A representative lot of this antibody bound BLAST-2/CD23 peptide with a KD of 1.7 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected BLAST-2/CD23 in Raji cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Syntaxin 4 in A431 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Syntaxin 4 in lysates from NIH3T3, and C6 cells and and Human kidney tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Syntaxin 4 in Human kidney and Human prostate tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Syntaxin 4 in C6 cells.

Target description

Low affinity immunoglobulin epsilon Fc receptor (UniProt: P06734; also known as BLAST-2, C-type lectin domain family 4 member J, Fc-epsilon-RII, Immunoglobulin E-binding factor, Lymphocyte IgE receptor, CD23) is encoded by the FCER2 (also known as CD23A, CLEC4J, FCE2, IGEBF) gene (Gene ID: 2208) in human. BLAST-2/CD23 is a homotrimeric, single-pass type II membrane glycoprotein with a cytoplasmic domain (aa 1-21), a transmembrane domain (aa 22-47), and an extracellular domain (aa 48-321). It can also exist as a soluble excreted form, sCD23 (aa 150-321), which is produced by ADAM10-mediated ectoderm shedding and has pleiotropic cytokine-like activities. BLAST-2/CD23 serves as a low-affinity receptor for immunoglobulin E (IgE) and CR2/CD21. It plays an essential role in the regulation of IgE production and in the differentiation of B-cells. Syntaxin-4 (UniProt: Q12846; also known as Renal carcinoma antigen NY-REN-31) is encoded by the STX4 (also known as STX4A) gene (Gene ID: 6810) in human. Syntaxins are a family of membrane proteins involved in vesicle trafficking. Syntaxin 4 (STX4) is single-pass type IV membrane protein with a single transmembrane domain (aa 276-296). It is a component of the SNARE complex that is composed of STX4, SNAP23 and VAMP7 that interacts with SYT7 during lysosomal exocytosis. In neuronal cells it localizes to neurite tips. It is recruited at neurite tips to membrane domains rich in the phospholipid 1-oleoyl-2-palmitoyl-PC that promotes neurite tip surface expression of the dopamine transporter SLC6A3/DAT by facilitating fusion of SLC6A3-containing transport vesicles with the plasma membrane. STX4 is reported to mediate retrograde signaling, modulating the membrane levels of synaptotagmin 4 and the trans-synaptic adhesion protein Neuroligin 1. STX4-dependent trafficking regulates synaptic development, including controlling synaptic bouton number and the ability to bud new varicosities in response to acute neuronal stimulation. Together with syntaxin binding protein 3 (STXB3) and vesicle-associated membrane protein 2 (VAMP2), it may also play a role in docking/fusion of intracellular GLUT4-containing vesicles with the cell surface in adipocytes. High expression of STX4 in skeletal muscle plays a critical role in insulin-stimulated glucose uptake by promoting translocation of GLUT4 to the cell surface. It is also involved in the regulation of glucose-stimulated insulin secretion in β cells. Two isoforms of STX4 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
