General description
Adhesion G protein-coupled receptor E4 (UniProt: Q91ZE5; also known as EGF-like module receptor 4, EGF-like module-containing mucin-like hormone receptor-like 4, F4/80-like-receptor, Seven-span membrane protein FIRE) is encoded by the Adgre4 (also known as Emr4) gene (Gene ID: 52614) in mouse. F4/80-like-receptor (Emr4), a member of the epidermal growth factor (EGF)-TM7 family, is a crucial glycoprotein predominantly expressed on murine macrophages, where it plays a significant role in immune regulation and macrophage biology. It is predominantly localized to the plasma membrane of macrophages, facilitating interactions with ligands and other immune cells. The protein is expressed in myeloid cells and also expressed on resident macrophages. Structurally, murine F4/80-like-receptor is a multipass membrane protein characterized by four extracellular domains, two EGF-like domains, four cytoplasmic domains, and seven-transmembrane domains, typical of G-protein-coupled receptors (GPCRs). This unique structure allows F4/80-like-receptor to mediate cell adhesion and signaling pathways essential for macrophage function. Additionally, research indicates that Emr4 may influence the pathogenesis of inflammatory diseases by regulating macrophage activation and function. Dysregulation of Emr4 expression has been linked to exacerbated inflammatory responses and could contribute to the progression of chronic inflammatory conditions. Specificity
Clone 1B19 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects mouse F4/80-like-receptor. It targets an epitope within 25 amino acids from the first extracellular domain.
Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the first extracellular domain of mouse F4/80-like-receptor.
Application
Quality Control Testing
Evaluated by Western Blotting in RAW 264.7 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected F4/80-like receptor in RAW 264.7 cell lysate.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound F4/80-like receptor peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected F4/80-like receptor in mouse spleen tissue sections.
Flow Cytometry Analysis: 1 µg from a representative lot detected F4/80-like receptor in thioglycolate-elicited mouse peritoneal macrophages.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
