Accurately measure out 100 mg of powdered turmeric extract and carefully transfer it to a 50 mL volumetric flask. Measure out 30 mL of acetone and use a sonicator for 30 minutes. Begin by marking the top with acetone, then mix and centrifuge. Transfer 5 mL of the supernatant to a 50 mL volumetric flask and fill it up to the mark with the mobile phase. Make sure to mix it thoroughly. Ensure proper filtration by using a 0.45 µm PVDF syringe filter before injecting into the HPLC system.

Description

General description

Turmeric is derived from the dried rhizome of Curcuma longa, a flowering plant belonging to the ginger family, Zingiberaceae. This plant is a perennial and is native to the Indian subcontinent and Southeast Asia. Turmeric possesses a warm and bitter flavor, reminiscent of black pepper, along with an earthy and mustard-like aroma. Curcumin, the main component of turmeric, has a vibrant yellow color. Curcumin has been recognized and approved as a food additive by reputable organizations such as the World Health Organization (WHO), European Parliament, and United States Food and Drug Administration (FDA).

Analysis Note

HPLC is used to determine the curcuminoids (curcumin, demethoxycurcumin and bisdemethoxycurcumin) in turmeric extract (powdered) following the USP monograph (Powdered Turmeric Extract; Dietary Supplements). The Purospher® Star RP-18 endcapped, 250 x 4.6 mm, 5 µm HPLC column is used to separate the curcuminoid peaks. System Suitability criteria (determined with solution of USP RS in mobile phase): Relative retention times are 1.0, 1.2, 1.4. Peak Tailing NMT 1.5 (observed: 1.04-1.06).Resolution NLT 2.0 between Curcumin and Desmethoxycurcumin (observed: 3.9).Resolution NLT 2.0 Between Desmethoxycurcumin and Bisdesmethoxycurcumin (observed: 3.8).RSD desmethoxycurcurmin NMT 2.0% (observed 0.14 %).
