Biochem/physiol Actions

当达到20-40°C时，ECM基质胶将发生热活化聚合，形成重组基底膜。 当PC12细胞在该ECM胶薄层上生长时，将在2天内形成神经突。

用于生产本品的每个小鼠种群都定期筛查病原体。经检测，LDEV呈阴性。

Preparation Note

该产品是从小鼠Engelbreth-Holm-Swarm肉瘤中提取的；并经过氯仿透析。 使用前，将凝胶置于2-8℃下解冻过夜，并使用预冷至2-8℃的板和移液器将凝胶分配至多孔板。该凝胶中可能含有不会影响其活性的沉淀物。 该凝胶也可使用2-8℃的Dulbecco′改良Eagle′培养基将凝胶稀释最多两倍，应先稀释、再分配至多孔板。 该产品在20°C下放置5分钟即可成胶。对于长时间操作，应在低于10°C的温度下进行。细胞可接种于0.5 mm厚的凝胶层上或在1mm厚的凝胶层内培养。 如果在凝胶层内培养细胞，请在接种前将细胞加至凝胶中，建议密度为3-4 x 104 个细胞/mL。 可使用溶剂为PBS，不含钙、镁或EDTA且浓度为0.6-2.4单位/mL的蛋白酶，将细胞从凝胶中分离出来。

Other Notes

ECM制剂的蛋白质浓度为8-12 mg/mL，主要成分为层粘连蛋白，还含有IV型胶原蛋白、蛋白多糖硫酸乙酰肝素、巢蛋白等其他次要成分。 向ECM凝胶中添加IV型胶原蛋白可增加其聚合度。
生长因子减少的修改可为培养基提供更多的控制，更好地解释结果并提高聚合度。 该培养基还缺少可能会干扰某些分析的酚红。
