一般描述

MSL3, also known as Male-specific lethal 3 homolog, or Male-specific lethal-3 homolog 1, or Male-specific lethal-3 protein-like 1 (MSL3-like 1), and encoded by the gene MSL3/MSL3L1, is a component of the MSL(male-specific lethal ) histone acetyltransferase complex which is responsible for the majority of histone H4 acetylation at ′Lys-16′ which is implicated in the formation of higher-order chromatin structure. MSL3 may also be involved in chromatin remodeling and transcriptional regulation. Additionally, MSL3 may have a role in X inactivation as its gene is a target for X inactivation. MSL3 is localized to the nucleus. MSL3 is expressed in many tissues including liver, pancreas, heart, lung, kidney, skeletal muscle, brain, and placenta, with highest expression in skeletal muscle and heart.
~58 kDa observed. Five isoforms are known to exist at 60 kDa (isoform 1), 53 kDa (isoform 2), 58 kDa (isoform 3), 40 kDa (isoform 4), and 48 kDa (isoform 5).

免疫原

Linear peptide corresponding to human MSL3-like 1.

应用

Chromatin Immunoprecipitation Analysis: A representative lot from an independent laboratory immunoprecipitated MSL3-like 1 from HeLa cell lysate (Wu, L., et al. (2011). Mol Cell. 43(1):132-144.).

Mammalian genomes harbor abundant transposable elements (TEs) and their remnants, with numerous epigenetic repression mechanisms enacted to silence TE transcription. However, TEs are upregulated during early development, neuronal lineage, and cancers, although the epigenetic factors contributing to the transcription of

Maintenance of genome integrity requires tight control of DNA damage response (DDR) signalling and repair, with phosphorylation and ubiquitination representing key elements. How these events are coordinated to achieve productive DNA repair remains elusive. Here we identify the ubiquitin-conjugating enzyme

Cancer is a complex disease manifestation. At its core, it remains a disease of abnormal cellular proliferation and inappropriate gene expression. In the early days, carcinogenesis was viewed simply as resulting from a collection of genetic mutations that altered the gene expression of key oncogenic genes or tumor suppressor genes leading to uncontrolled growth and disease (Virani, S et al 2012). Today, however, research is showing that carcinogenesis results from the successive accumulation of heritable genetic and epigenetic changes. Moreover, the success in how we predict, treat and overcome cancer will likely involve not only understanding the consequences of direct genetic changes that can cause cancer, but also how the epigenetic and environmental changes cause cancer (Johnson C et al 2015; Waldmann T et al 2013). Epigenetics is the study of heritable gene expression as it relates to changes in DNA structure that are not tied to changes in DNA sequence but, instead, are tied to how the nucleic acid material is read or processed via the myriad of protein-protein, protein-nucleic acid, and nucleic acid-nucleic acid interactions that ultimately manifest themselves into a specific expression phenotype (Ngai SC et al 2012, Johnson C et al 2015). This review will discuss some of the principal aspects of epigenetic research and how they relate to our current understanding of carcinogenesis. Because epigenetics affects phenotype and changes in epigenetics are thought to be key to environmental adaptability and thus may in fact be reversed or manipulated, understanding the integration of experimental and epidemiologic science surrounding cancer and its many manifestations should lead to more effective cancer prognostics as well as treatments (Virani S et al 2012).
