General description
Yellow fever virus is an arbovirus of the Flavivirus genus. It is a single-stranded RNA virus, characterized by its spherical shape and an envelope derived from host cell membranes. The viral genome encodes a polyprotein (UniProt: P03314, Gene ID: 1502173) in Yellow fever virus (strain 17D), that is cleaved into structural proteins, including the capsid (C), surrounded by a lipid bilayer derived from the host cell, which contains two key envelope proteins: pre-membrane (prM), and envelope (E) proteins, essential for viral assembly and entry into host cells. The Envelope protein E plays a critical role in mediating fusion with host cell membranes, and its structure undergoes conformational changes upon exposure to neutral pH, facilitating viral entry. Clone 2D12.A is a mouse monoclonal antibody that specifically reacts with the envelope protein of the wild (Asibi) and vaccine strains of yellow fever virus. YFV is the cause of acute viral hemorrhagic disease transmitted by mosquitoes. YFV primarily infects hepatocytes, where it replicates and leads to cell death, contributing to the pathogenesis of yellow fever, a severe hemorrhagic disease fever, including fever, chills, loss of appetite, fatigue and in severe cases, liver failure, hemorrhage, and even death. Preparation Note
0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
