General description

Runt-related transcription factor 1 (UniProt Q01196; also known as Acute myeloid leukemia 1 protein, CBF-alpha-2, Core-binding factor subunit alpha-2, Oncogene AML-1, PEA2-alpha B, PEBP2-alpha B, Polyomavirus enhancer-binding protein 2 alpha B subunit, SL3-3 enhancer factor 1 alpha B subunit, SL3/AKV core-binding factor alpha B subunit) is encoded by the RUNX1 (also known as AML1, CBFA2) gene (Gene ID 861) in human. RUNX1 is a runt-related transcription factor family member that plays a role in the differentiation of hematopoietic stem cells into mature blood cells. RUNX1 forms a core-binding factor (CBF) complex with core binding factor β (CBFβ; encoded by the CBFB gene), and both RUNX1 and CBFB genes are well known targets of chromosomal translocations in acute myeloid leukemia (AML), generating the leukemogenic fusion proteins AML1-ETO and CBFB-MYH11. RUNX1 contains an N-terminal Runt homology domain (RHD; a.a. 50-178) that mediates protein-protein interaction, followed by a DNA-binding region (a.a. 80-177) that mediates the binding of the core consensus sequence TGTGGNNN (where NNN can represent either TTT or TCA). RUNX1 can bind DNA as a monomer, but its DNA-binding affinity is enhanced by 10-fold when complexed with CBFβ.

Target band(s) size can vary depending on phosphorylation and isoform(s) present in sample(s). 50.70 kDa (soform AML-1A), 48.74 kDa (AML-1B), 27.43 kDa (AML-1C), 28.23 kDa (AML-1E), 51.82 kDa (AML-1G), 49.08 kDa (AML-1H), 50.17 kDa (AML-1I), 37.73 kDa (AML-1L; AML1-delta N) calculated. Uncharacterized band(s) may appear in some lysates.

Epitope: Region between DNA-binding domain and KAT6A-binding domain.
GST-tagged recombinant fragment from the region between the DNA-binding domain and the KAT6A-binding domain of human RUNX1.

Western Blotting Analysis: A 1:500 dilution from a representative lot detected RUNX1 in 10 µg of human TF-1 erythroleukemic cell and murine RAW264.7 macrophage lysates.

Biochem/physiol Actions

Expected to react with isoform AML-1A, AML-1B, AML-1E, AML-1G, AML-1H, AML-1I, AML-1L (AML1-delta N), but not AML-1FA, AML-1FB, or AML-1FC. Reactivity toward isoform AML-1C is likely, but has not been confirmed.

Preparation Note

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Evaluated by Western Blotting Analysis in K562 cell lysate:

Western Blotting Analysis: A 1:500 dilution of this antibody detected RUNX1 in 10 µg of human K562 erythroleukemic cell lysate.
