General description
Growth hormone secretagogue receptor type 1 (UniProt: Q92847; also known as Ghrelin Receptor, GHS-R, GH-releasing peptide receptor) is encoded by the GHSR gene (Gene ID: 2693) in human. The ghrelin receptor is a G protein-coupled receptor (GPCR) primarily involved in the regulation of energy homeostasis and growth hormone release. The receptor is characterized by its seven-transmembrane domain structure, typical of GPCRs. Ghrelin, a peptide hormone, acquires its biological activity through octanoylation at the third serine residue, and the ghrelin receptor accommodates this acylated peptide with its bifurcated characteristic ligand-binding pocket. Since the ghrelin receptor is predominantly expressed in the hypothalamus and pituitary gland, ghrelin plays a crucial role in stimulating growth hormone secretion. Additionally, it is found in peripheral tissues such as the heart and gastrointestinal tract, suggesting its involvement in broader physiological processes considering its ligand, Ghrelin has a wide range of physiological functions, playing roles in growth hormone secretion, adiposity, energy homeostasis, memory formation, and hippocampal neurogenesis. The Ghrelin Receptor can also form heterodimers with other receptors, which may alter its signaling pathways and functional outcomes. Dysregulation of Ghrelin signaling through Ghrelin Receptor is associated with obesity, cachexia, and certain cardiovascular diseases. Ghrelin/GHSR axis also plays a role in promoting tumor growth and progression. GHSR and Ghrelin expression is more common in high grade central nervous system (CNS) tumors that may increase the growth of gliomas through an autocrine/paracrine mechanism.
