Cadherin 17 (UniProt: Q12864; also known as Intestinal peptide-associated transporter HPT-1, Liver-intestine cadherin, LI-cadherin) is encoded by the CDH17 gene (Gene ID: 1015) in human. Cadherins are calcium-dependent cell adhesion molecules that participate in cell-cell adhesion during embryogenesis, development, organogenesis, and differentiation. Cadherin 17 is a single-pass type I membrane glycoprotein. It is predominantly expressed in the gastrointestinal tract and pancreatic duct and is not detected in kidney, lung, liver, brain, adrenal gland, and skin. It is synthesized with a signal peptide (aa 1-22), which is cleaved off to generate the mature form that contains an extracellular domain (aa 23-787), transmembrane domain (aa 788-808), and a cytoplasmic domain (aa 809-832). It differs from classical cadherins in having a short cytoplasmic tail at its carboxyl-terminus. In the extracellular region it has 7 cadherin domains. Three calcium ions are usually bound at the interface of each cadherin domain that rigidify the connections, imparting a strong curvature to the full-length ectodomain. Cadherin-17 expression has been described in several gastric carcinomas and it is shown to be a good immunohistochemical marker for diagnosis of adenocarcinomas of the digestive system. It is considered as an oncofetal molecule associated with poor prognostic outcomes of hepatocellular carcinoma. (Ref.: Wang, Y., et al. (2013). PLoS One. 8(9); e72386).
