General description
Cyclic guanosine monophosphate (cGMP)-specific 3′, 5′-cyclic phosphodiesterase (PDE5A) is an 875-amino acid protein. It is expressed in platelets and smooth muscle cells. PDE5A is a homodimer with two regulatory domains that is, a carboxyl-terminal catalytic domain and an amino-terminal domain containing two allosteric cGMP-binding sites. The gene encoding this protein is present on chromosome 4q25-27.

Application

Biochem/physiol Actions
Cyclic guanosine monophosphate (cGMP)-specific 3′, 5′-cyclic phosphodiesterase (PDE5A) is involved in the regulation of nitric-oxide-generated cGMP, smooth muscle relaxation and neuronal survival. It hydrolyzes cGMP to 5′-GMP. PDE5A is involved in pain signaling during migraine-like attacks. PDE5A has been shown to be overexpressed in squamous and transitional cell carcinomas.
