General description
Cell adhesion molecule-related, downregulated by oncogenes (CDON) gene is mapped to human chromosome 11q23–24. The CDO protein encoded by this gene is a known cell surface receptor of the immunoglobulin superfamily This cell surface protein consists of five Ig repeats and three fibronectin type III (FNIII) repeats in its extracellular region, a transmembrane domain and a peculiar 270 amino acid intracellular region.

Application

Biochem/physiol Actions
Cell adhesion molecule-related, downregulated by oncogenes (CDON) acts as a tumor suppressor gene in neuroblastomas. It acts as a high potential therapeutic target for PCa (prostate cancer). CDON functions as a receptor in SHH (sonic hedgehog) signaling pathway involved in neural crest cell fate during embryonic development. It is also known to activate apoptotic process in the absence of SHH pathway. CDO protein enhances myocardium formation of pluripotent stem cells.
Nonsense mutation of CDON gene involved in SHH signaling will lead to congenital defect called as holoprosencephaly (HPE).
CDO associates with co-receptor BOC (brother of CDO[cell adhesion molecule, down-regulated by oncogenes)] by mutually linking their internal domains and ectodomains. This complex is involved in the regulation of myogenic differentiation.
