General description
The gene FAM83G (family with sequence similarity 83 member G), also referred to as PAWS1 (protein associated with SMAD1), is found to be conserved across vertebrates and is a member of a family of hypothetical proteins, FAM83A–H. These proteins contain a conserved N-terminal domain called DUF1669, whose function is yet to be determined. This domain contains a putative pseudo-phospholipase D motif.

Biochem/physiol Actions
The gene FAM83G (family with sequence similarity 83 member G) encodes a protein that interacts with SMAD1 (similar to mothers against decapentaplegic 1). Independent of SMAD4, the encoded protein forms a complex with SMAD1 and forms a substrate for type I BMP (bone morphogenetic protein) receptor kinases. PAWS1 also forms a substrate for ALK3 (aurora-like kinase ), a type IA BMP receptor, and its phosphorylation is induced by BMPs. Phosphorylated PAWS1 regulates the expression of NEDD9 (neural precursor cell expressed, developmentally down-regulated 9 ) and ASNS (asparagine synthetase), which are SMAD4-independent BMP target genes. It also regulates the expression of several genes independent of BMP stimulation. PAWS1 is involved in the regulation of BMP pathway.
