General description

p38-interacting protein (p38IP), also named family with sequence similarity 48, member A (FAM48A) p38IP comprises nuclear localization sequence, two serine rich domains and a PEST domain. It is localized in the nucleus and the cytoplasm. It partially colocalizes with autophagy related protein 9 (Atg9). p38IP is mapped to human chromosome 13q13.3.

Immunogen

synthetic peptide corresponding to an internal region of human FAM48A, conjugated to KLH. The corresponding sequence is identical in mouse, rat, bovine, canine and monkey.

Application

Anti-FAM48A /P38IP antibody produced in rabbit may be used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

Anti-FAM48A /P38IP recognizes human and monkey FAM48A /P38IP.
p38-interacting protein (p38IP) binds directly to p38 mitogen activated protein kinase and is required for its activation in vivo. Both p38 and p38IP were found to be necessary for E-cadherin down-regulation during gastrulation. Recently, p38IP was identified as an interacting protein for autophagy-related protein 9 (mAtg9), a transmembrane protein required for autophagy. The interaction of p38IP with mAtg9 is regulated by p38. p38IP is required for starvation-induced mAtg9 trafficking and autophagosome formation.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Preparation Note

For continuous use, store at 2-8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
