一般描述
Angiopoietins are a family of growth factors that bind endothelial receptor tyrosine kinase Tie2. While Ang1 and Ang2 act as antiagonist, Ang3 and Ang4 have been shown as agonists to Tie2.
Angiopoietins are growth factors vital for vascular differentiation during normal development. There are four angiopoietins discovered that bind to receptor tyrosine kinases Tie2. Human Angiopoietin-4 (ANG-4) expression is observed in lung and cultures HUVEC cells. Angiopoietin-4 mediates angiogenesis and migration of cells and is overexpressed in colorectal and gastric adenocarcinomas and glioblastomas. The cellular functions of ANG-4 are mediated by Akt and ERK1/2 pathways. The roles of ANG-3 and ANG-4 are significant in the survival of tumor cells, enhance angiogenesis and increase vascular branching. Reports also show that ANG-4 inhibits the angiogenesis in human small cell lung cancer cell line . Anti-Angiopoietin-4 detects human angiopoietin-4. The antibody shows less than 1% cross reactivity with recombinant human ANG-1 and recombinant human ANG-2 in ELISA.
免疫原
purified NSO-derived recombinant human ANG-4.
应用
Anti-Angiopoietin-4 antibody may be used for immunoblotting at a working concentration of 0.1-0.2 μg/ml. For ELISA, the recommended concentration is 0.5-1.0 μg/ml. Antibody concentration of 10 μg/ml may be used for immunohistochemistry of cells and tissues in chromogenic detection system. For blocking assays, antibody concentration of 1-3 μg/ml would block 50% of recombinant human ANG-4. At 50 μg/ml approximately 50% blocking may be achieved.

Angiopoietin-4 promotes glioblastoma progression by enhancing tumor cell viability and angiogenesis.
Melissa K Brunckhorst et al.
Cancer research, 70(18), 7283-7293 (2010-09-09)
Glioblastoma multiforme (GBM) is a highly invasive and vascularized aggressive brain tumor. Less than 10% of GBM patients survive >5 years after diagnosis. Angiogenesis plays an important role in GBM growth, and antiangiogenesis-based therapies have shown clinical efficacy for GBM
Minna W B Olsen et al.
Neoplasia (New York, N.Y.), 8(5), 364-372 (2006-06-23)
Angiopoietins (Ang) are involved in the remodeling, maturation, and stabilization of the vascular network. Ang-4 was discovered more recently; thus, its effect on angiogenesis and its interplay with other angiogenic factors have not been equivocally established. The role of Ang-4

The angiopoietins have recently joined the members of the vascular endothelial growth factor family as the only known growth factors largely specific for vascular endothelium. The angiopoietins include a naturally occurring agonist, angiopoietin-1, as well as a naturally occurring antagonist

The angiopoietin (Ang) family of growth factors includes Ang1, Ang2, Ang3, and Ang4, all of which bind to the endothelial receptor tyrosine kinase Tie2. Ang3 (mouse) and Ang4 (human) are interspecies orthologs. In experiments with human endothelial cell lines, Ang3
