General description
Angiopoietins are a family of growth factors that bind endothelial receptor tyrosine kinase Tie2. While Ang1 and Ang2 act as antiagonist, Ang3 and Ang4 have been shown as agonists to Tie2.
Angiopoietins are growth factors vital for vascular differentiation during normal development. There are four angiopoietins discovered that bind to receptor tyrosine kinases Tie2. Human Angiopoietin-4 (ANG-4) expression is observed in lung and cultures HUVEC cells. Angiopoietin-4 mediates angiogenesis and migration of cells and is overexpressed in colorectal and gastric adenocarcinomas and glioblastomas. The cellular functions of ANG-4 are mediated by Akt and ERK1/2 pathways. The roles of ANG-3 and ANG-4 are significant in the survival of tumor cells, enhance angiogenesis and increase vascular branching. Reports also show that ANG-4 inhibits the angiogenesis in human small cell lung cancer cell line . Anti-Angiopoietin-4 detects human angiopoietin-4. The antibody shows less than 1% cross reactivity with recombinant human ANG-1 and recombinant human ANG-2 in ELISA.

purified NSO-derived recombinant human ANG-4.
Application
Anti-Angiopoietin-4 antibody may be used for immunoblotting at a working concentration of 0.1-0.2 μg/ml. For ELISA, the recommended concentration is 0.5-1.0 μg/ml. Antibody concentration of 10 μg/ml may be used for immunohistochemistry of cells and tissues in chromogenic detection system. For blocking assays, antibody concentration of 1-3 μg/ml would block 50% of recombinant human ANG-4. At 50 μg/ml approximately 50% blocking may be achieved.
