一般描述
α-淀粉酶主要在唾液腺和胰腺中大量产生。在空肠和乳腺中也可表达。编码α淀粉酶的基因是位于1号染色体短臂上的AMY1和AMY2。AMY1负责在唾液和乳腺中产生这种酶,而AMY2则在胰腺中合成这种酶。AMY1也在某些肿瘤组织中表达。哺乳动物淀粉酶由三个结构域(A、B和C)组成。结构域A包含具有两个天冬氨酸和一个谷氨酸残基的活性位点。结构域B毗邻活性位点区,它是维持蛋白质构象是必不可少的结构。结构域C涉及催化机理。

应用
兔源抗α-淀粉酶抗体可用于免疫荧光和免疫组织化学分析。
兔源抗α淀粉酶抗体还被用于免疫细胞化学分析。
将小鼠胰腺切片固定在4%多聚甲醛中,并使用兔抗淀粉酶抗体以1:100的稀释度用于免疫组织化学。
生化/生理作用
α--淀粉酶对于催化淀粉消化的第一步是必不可少的,淀粉消化是人类饮食中碳水化合物的主要来源。α-淀粉酶水解多糖,最终生成的主要产物为麦芽糖、麦芽三糖和极限糊精。
该抗血清对在人唾液和人胰腺提取物中发现的人α-淀粉酶具有特异性。未观察到与其他人唾液蛋白或胰腺提取蛋白的反应。
