General description

Crk proteins belong to a family of adaptor proteins with distinct Src homology (SH) domains, SH2 and SH3. Crk-I, Crk-II and CrkL are the cellular homologs of oncogene v-Crk of avian sarcoma virus. While Crk-I and Crk-II are the products of alternate gene splicing, Crk-L is encoded by a different gene although it shares 60% homology with Crk-II. The Crk proteins are involved in many signal transduction pathways. The SH2 domain of Crk proteins interact with several tyrosine kinases such as EGFR, Cbl, p130cas, paxillin and IRS-1. In humans, both SH2 and SH3 domains of Crk proteins are essential for neuronal differentiation of PC-12 cells, play active role in NGF and p21ras signalling.
Anti-Crk-II specifically recognizes human and chicken Crk-II (38 kDa).

Immunogen

synthetic peptide K-THVRLLDQQNPDEDFS corresponding to the C-terminal of human Crk-II (amino acids 289-304 with N-terminally added lysine), conjugated to KLH. The sequence is identical in human, mouse and chicken Crk-II and highly conserved (single amino acid substitution) in rat Crk-II. This sequence is absent in Crk-I isoform and has limited homology (60%) to Crk-L..

Application

Anti-Crk-II antibody may be used at a working dilution of 1:5000 in whole extract of chicken fibroblasts. It may be used for detection of Crk-II in whole extract of Raji cell line (Burkitt lymphoma). It is also suitable for protein microarray.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
