应用

来自球形节杆菌的胆碱氧化酶已被用于:
- 酶法测定胆碱(Cho)和磷酸胆碱(P-Cho)。
- 在鞘磷脂试验中通过测量游离胆碱来测定鞘磷脂的浓度
- 用于电化学测量的全组装记录设备的酶涂层
生化/生理作用
胆碱氧化酶催化胆碱到甘氨酸甜菜碱的四电子氧化反应,其中甜菜碱醛为中间体,分子氧为初级电子受体。该酶还可以甜菜碱醛作为底物。这可研究胆碱转化成醛中间体和甜菜碱醛转化成甘氨酸甜菜碱的反应机理。该酶是分子量约为83,000 Da(凝胶过滤法)或71,000 Da(SDS凝胶电泳法)的黄素蛋白。最佳pH约为pH 7.5,等电点(pI)约为pH 4.5。
其他说明
在37℃、pH 8.0条件下,一单位酶每分钟可形成1 μmM H2O2 ,同时将1 μmM胆碱氧化成甜菜碱醛。注意:在胆碱氧化酶将胆碱转化成甜菜碱过程中,每μM胆碱产生2 μmM H2O2 。
