Mast cells play an important role in innate immunity or the response of the body to invading pathogens. The functions of mast cells are regulated by a number of surface receptors; c-Kit (Stem cell factor receptor, SCF R; CD117) being the most critical. The signalling triggered by c-Kit results in mast cell activation in response to antigens. The response involves tyrosine phosphorylation by specific tyrosine kinases that drive the downstream signalling process. The secondary pathways activated by c-Kit include Akt, mTOR, MAPK, Sos-Grb2 and c-Jun. Phosphorylation of tyrosine 730 is required for the binding of PLCγ leading to activation of various signalling pathways In addition to development of mast cells, c-Kit also regulates their subsequent maturation and homing to the residence sites. Constitutive activation by SCF R is implicated in myeloproliferative discorders, systemic mastocytosis and gastrointestinal stomal cell tumors
