Biochem/physiol Actions

Preparation Note
该产品是从小鼠Engelbreth-Holm-Swarm肉瘤中提取的;并经过氯仿透析。 使用前,将凝胶置于2-8℃下解冻过夜,并使用预冷至2-8℃的板和移液器将凝胶分配至多孔板。该凝胶中可能含有不会影响其活性的沉淀物。 该凝胶也可使用2-8℃的Dulbecco′改良Eagle′培养基将凝胶稀释最多两倍,应先稀释、再分配至多孔板。 该产品在20°C下放置5分钟即可成胶。对于长时间操作,应在低于10°C的温度下进行。细胞可接种于0.5 mm厚的凝胶层上或在1mm厚的凝胶层内培养。 如果在凝胶层内培养细胞,请在接种前将细胞加至凝胶中,建议密度为3-4 x 104 个细胞/mL。 可使用溶剂为PBS,不含钙、镁或EDTA且浓度为0.6-2.4单位/mL的蛋白酶,将细胞从凝胶中分离出来。
Other Notes
