FER is a nonreceptor protein-tyrosine kinase (PTK), which has a ubiquitous expression pattern. Along with Fps/Fes protein, it makes up the subfamily called PTK. It contains three predicted coiled-coil (CC) motifs in its N-terminal, a Src homology 2 (SH2) domain located in the center, and a catalytic domain at its C-terminal. An isoform of this protein is specific to the testis, and is called FerT.
生化/生理作用
In fibroblasts, FER acts downstream of epidermal growth factor (EGF) and platelet-derived growth factor (PDGF) receptors. It is also activated downstream of FcγRI, a high-affinity receptor for immunoglobulin E (IgE), in mast cells. However, the exact role in these pathways is yet unknown. Inactivation of this protein in mice results in reduction in cortactin phosphorylation, though they are viable and fertile. Thus, Fer might be involved in controlling cell growth, adhesion, development of retina and spermatogenesis. It is essential for the phosphorylation of cortactin.
