General description

FER is a nonreceptor protein-tyrosine kinase (PTK), which has a ubiquitous expression pattern. Along with Fps/Fes protein, it makes up the subfamily called PTK. It contains three predicted coiled-coil (CC) motifs in its N-terminal, a Src homology 2 (SH2) domain located in the center, and a catalytic domain at its C-terminal. An isoform of this protein is specific to the testis, and is called FerT.

Biochem/physiol Actions

In fibroblasts, FER acts downstream of epidermal growth factor (EGF) and platelet-derived growth factor (PDGF) receptors. It is also activated downstream of FcγRI, a high-affinity receptor for immunoglobulin E (IgE), in mast cells. However, the exact role in these pathways is yet unknown. Inactivation of this protein in mice results in reduction in cortactin phosphorylation, though they are viable and fertile. Thus, Fer might be involved in controlling cell growth, adhesion, development of retina and spermatogenesis. It is essential for the phosphorylation of cortactin.

Physical form

Supplied in 50 mM Tris-HCl, pH 7.5, with 150 mM NaCl, 0.2 5mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF, and 25% glycerol.

Fer is a nuclear and cytoplasmic intracellular tyrosine kinase. Herein we show that Fer is required for cell-cycle progression in malignant cells. Decreasing the level of Fer using the RNA interference (RNAi) approach impeded the proliferation of prostate and breast

Fps/Fes and Fer are the only known members of a distinct subfamily of the non-receptor protein-tyrosine kinase family. Recent studies indicate that these kinases have roles in regulating cytoskeletal rearrangements and inside out signalling that accompany receptor ligand, cell matrix

The ubiquitous Fer protein-tyrosine kinase has been proposed to regulate diverse processes such as cell growth, cell adhesion, and neurite outgrowth. To gain insight into the biological function of Fer, we have targeted the fer locus with a kinase-inactivating missense
