General description

Protein-tyrosine phosphatase receptor type J (PTPRJ/CD148) is present predominatly in epithelial cells, various tumor cell lines and in lymphocytes. PTPRJ is commonly referred to as CD148 or DEP1.

Biochem/physiol Actions

Protein-tyrosine phosphatase receptor type J (PTPRJ/CD148) is a protein tyrosine phosphatase. It is a negative regulator of T-cell receptor (TCR)-mediated activation of T cells. CD148 affects TCR signaling by interfering with the phosphorylation of phospholipase Cγ1 (PLCγ1) and LAT. CD148 dephosphorylates platelet-derived growth factor β-receptor, resulting in reduction in cell proliferation. CD148 also participates in functioning of cell-cell contacts and adherens junctions. It interacts with p120(ctn), a component of adherens junctions. CD148 associates with receptor tyrosine kinase RET and modulates the tumor phenotype associated with chimeric RET/papillary thyroid carcinoma oncogenes. It dephosphorylates p85 regulatory subunit of phosphoinositide 3-kinase (PI3K) and ERK1/2 (Extracellular signal-regulated kinases). It dephosphorylates and stabilizes epidermal growth factor receptor (EGFR). CD148 negatively regulates Fms-like tyrosine kinase-3 (FLT3) phosphorylation and signaling. It is up-regulated in humans suffering from rheumatoid arthritis and down-regulated in breast tumors. CD148 is a positive regulator of platelet activation and thrombosis.
