一般描述
脂氧化酶(LOX)又名脂氧合酶,是一种非血红素铁蛋白。脂氧合酶在植物、部分微生物和动物中广泛存在。根据pH值和最适温度、底物特异性以及不同试剂的抑制程度,分为LOX-1、LOX-2和LOX-3。
应用
大豆酶将使用花生四烯酸作为底物,以亚油酸作为底物的活性约为 15%;花生四烯酸氧化的产物是 12-或 15-过氧化氢花生四烯酸(12-HPETE 或 15-HPETE)。
来自大豆的脂氧合酶已用于:
- 研究2-(α-D-吡喃葡萄糖基)甲基-2,5,7,8-四甲基色胺-6-醇(TMG)对脂蛋白氧化的影响
- 作为脂氧合酶的代表酶,在微孔板检测平台上筛选木材提取物降解酶
- 作为脂氧合酶抑制活性检测的阳性对照品
生化/生理作用
催化含有顺式,顺式-1,4-戊二烯结构的脂质的氢过氧化。
在接近100℃的温度下热处理5到10分钟,使脂氧合酶失活。研究表明,γ辐射处理降低了大豆脂氧合酶的比活性。这类酶可用于与哮喘和过敏相关的研究。脂氧合酶适用于工业生物转化,如芳香精细化学品的生产。

当亚油酸为 3.0 mL 体积(1 cm 光径)底物时,25°C、pH 9.0 条件下 1 个单位每分钟将导致 A 234 增加 0.001。一个 A 234 单位相当于氧化 0.12μ 摩尔亚油酸。
