一般描述

天然完整的人纤溶酶原是791个氨基酸的糖蛋白,具有多达 24个二硫键。纤溶酶原包含单个N-连接的唾液酸化双触角聚糖。两个O-聚糖具有Gal β-1-3GalNAc 核心,其在末端Gal 处是α-2-3唾液酸化的。另外的二唾液酸化形式具有第二唾液酸残基,其与GalNAc 具有α-2-6键。单 -和二唾液酸化形式在人纤溶酶原中以 80:20的摩尔比发生。

生化/生理作用

纤溶酶原是蛋白酶纤溶酶的无活性前体。纤溶酶原由组织型纤溶酶原激活剂 (tPA) 激活,tPA 主要激活纤溶酶的纤维蛋白溶解(血栓溶解)活性,或者由尿激酶型纤溶酶原激活剂 (uPA) 激活,uPA 与细胞外基质重塑和细胞迁移有关。纤溶酶可裂解纤维蛋白/纤维蛋白原和凝血因子 V/Va 以及 VIII/VIIIa。它可通过裂解非活性酶原激活基质金属蛋白酶。还与某些生长因子的激活有关,如血管內皮生长因子 (VEGF) 和转化生长因子 β (TGF-β)。

制备说明

溶液应储存在含 20mM 赖氨酸的中性 pH 缓冲液中。蛋白质应尽量浓缩。分成小份在 -20°C 下冻存,可稳定保存几周。

分析说明

不含 ε-氨基己酸
经检测,每位捐献者的血浆 HIV-1/HIV-2 抗体、HCV 和 HbSAg 抗体均为阴性。

其他说明

当测定 5.0mL 体积中的高氯酸可溶性产物时,在 pH 7.5 和 37°C 下,一单位本品将使 α-酪蛋白在 20 分钟内产生 1.0 的 ΔA275。活性在尿激酶激活纤溶酶后测定。
