General description

Stromal cell-derived factor-1 (SDF-1) is a proinflammatory cytokine that belongs to the CXC family of chemokines. SDF-1 exists as two isoforms, SDF-1β and SDF-1α. The effects of SDF-1 are mediated by its binding to the receptor CXCR4, and sometimes to CXCR7. Both lymphoid (bone marrow, spleen and thymus) and non-lymphoid (stomach, kidney, liver, lung, brain, heart) organs express SDF-1. The most important functions assigned to SDF-1 are angiogenesis, migration of μglia and astrocytes, tissue repair and as chemoattractant to hematopoietic cells. Studies also implicate SDF-1 in survival of human glioma cells via the activation of PI3K/Akt pathway. The other pathways that are activated by SDF-1 include MAPK, NF-κB, paxillin, Crk and other focal adhesion proteins. A variety of human malignancies of pancreas, breast, prostate and glioma exhibit overexpression of CXCR4. High expression of SDF-1 has been observed in lymph nodes, lungs, bone and liver that are the most common sites of metastasis of breast cancer cells. Anti- SDF-1β/PBSF recognizes recombinant human SDF-1β/PBSF. By immunoblotting, this antibody exhibits less than 5% cross-reactivity with recombinant human SDF-1α.

recombinant human SDF-1β/PBSF expressed in E. coli.

Application

Anti-SDF-1β/PBSF antibody may be used for immunoblotting at a working concentration of 0.1-0.2 μg/ml. For ELISA, the recommended working antibody concentration is 0.5-1.0 μg/ml. The antibody is also suitable for neutralization assays; ND50 of the antibody is ~5-15 μg/ml.
