General description
Monoclonal Anti-PTPA/ PPP2R4 (mouse IgG2a isotype) is derived from the hybridoma 5G3 produced by the fusion of mouse myeloma cells and splenocytes from the immunized BALB/c mice. Phosphotyrosyl phosphatase activator (PTPA), also known as protein phosphatase 2A activator, regulatory subunit 4 (PPP2R4), is a conserved protein from yeast to human. PTPA belongs to the B′ family and encodes a specific phosphotyrosyl phosphatase activator of the dimeric form of protein phosphatase 2A. The PTPA gene is mapped to human chromosome 9q34. Alternative splicing results in multiple transcript variants encoding different isoforms.
Immunogen
recombinant mouse PPP2R4, fusion protein
Application
Anti-PTPA/ PPP2R4 antibody, Mouse monoclonal is suitable for immunoblotting.
Monoclonal Anti-PTPA/ PPP2R4 antibody is suitable for use in western blot (0.5-1.0 μg/mL using whole extracts of mouse 3T3 or human HeLa cells).
Biochem/physiol Actions
Monoclonal Anti-PTPA/ PPP2R4 recognizes mouse and human PTPA/PPP2R4.
Phosphotyrosyl phosphatase activator (PTPA) is essential for the biogenesis of specific and active protein phosphatase 2A (PP2A). This protein acts as a peptidyl prolyl isomerase. It is known to promote the tyrosyl phosphatase activity of protein phosphatase 2A core heterodimer (PP2AD). PTPA regulates the PP2A-like enzymes. Deletion of the gene is considered lethal.
Protein phosphatase 2 phosphatase Activator (PTPA) is a protein phosphatase 2A (PP2A)-binding protein that activates the tyrosyl phosphatase function of PP2A and decreases the phosphoserine phosphatase function. Increased expression of PTPA has been linked to caspase-3 dependent cell death .
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discardedif not used within 12 hours.
