General description

Monoclonal Anti-KISS1 (mouse IgG1 isotype) is derived from the hybridoma KISS-54 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a peptide. Kisspeptin is the ligand for G-protein coupled receptor 54 (GPR54). It is expressed in the placenta, hypothalamus and basal ganglia. Kisspeptins are encoded by the gene KISS1 located on the human chromosome at 1q32.1. This protein belongs to the family of arginine-phenylalanine (RF) protein hormones. KISS1 gene is expressed in both central and peripheral tissues.

Immunogen

peptide corresponding to a sequence at the C-terminus of human KISS1, conjugated to KLH. The isotype is determined using a double diffusion immunoassay using Mouse Monoclonal Antibody Isotyping Reagents (Sigma ISO-2).

Application

Monoclonal Anti-KISS1 antibody produced in mouse may be used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

Monoclonal Anti- KISS1 recognizes human, bovine and canine KISS1.
KISS1 gene is considered as a human metastasis suppressor gene that can suppress melanoma and breast cancer metastasis, without affecting tumorigenicity. Kisspeptin modulates events downstream of cell-matrix adhesion in mechanisms involving cytoskeletal reorganization. Kisspeptin is involved in initiating the secretion of gonadotropin-releasing-hormone (GnRH) at puberty. This protein functions in the regulation of the hypothalamic-pituitary-gonadal axis, resulting in, influencing fertility and reproduction. KISS1 protein has been suggested as a possible treatment for reproductive disorders as well as clinical treatment of metastases.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Preparation Note

For extended storage, freeze at -20 °Cinworking aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discardedif not used within 12 hours.
