3-BrOP 是糖酵解抑制剂 3-溴丙酮酸 (3-BrPA) 的一种细胞渗透性酯,已知可靶向己糖激酶 II (HK2) 和 GAPDH。3-BrOP 进入细胞后被细胞内酯酶水解为 3-BrPA。3-BrOP 与 mTOR 抑制剂雷帕霉素具有协同杀伤肿瘤细胞的作用(雷帕霉素 100 ng/mL/mL 诱导肿瘤细胞凋亡 = 64%/36%Raji 人淋巴瘤细胞 + 40 μmol/l 雷帕霉素 40 μmol/l)。M 3-BrOP 和 72%/45% 的 HL-60 白血病细胞经 20μM 3-BrOP in 24 h)由于细胞 ATP 耗竭和葡萄糖摄取抑制增强。通过尾静脉注射给药,3-BrOP 可显著抑制 SK-N-SH 异种移植物来源的神经母细胞瘤在小鼠体内的肿瘤生长(14 天内增长 78%;20 mg/kg/天)。
