Endogenous galanin receptor GAL2/3 agonist with anti-obesity efficacy in cultures and in vivo.
Spexin 1 (SPX) is a 14-aa amidated peptide that acts as an endogenous galanin receptor GAL2/3 agonist (EC50 = 161 nM/GAL2, 626 nM/GAL3, >10 µM/GAL1 by reporter assays using respective HEK293 transfectants) that inhibits facilitated fatty acid (FA) uptake in diet-induced obesity (DIO) mouse adipocytes cultures (0.1-1 ng/mL) and induces rat stomach muscle contraction ex vivo (EC50 = 750 nM). SPX reduces caloric intake in rats (∼32%; 35 µg/kg/day s.c.) with corresponding weight loss and significantly reduces the RER at night as well as increases locomotion in mice in vivo (25 µg/kg/day i.p.). Note: peptide sequence is conserved in human, murine, rat and many other mammalian species.
