General description

BMPs (bone morphogenetic proteins) belong to the TGF-β superfamily of structurally related signaling proteins. As implied by their name, BMPs promote and regulate bone development, growth, remodeling and repair, in both prenatal development and postnatal growth of eye, heart, kidney, skin, and other tissues. BMP-10 is essential for the proper development of the heart, but is not expressed until after cardiac patterning or looping are completed. Recombinant human BMP-10 is a 32 kDa disulfide-linked homodimer containing two 142 amino acid polypeptide chains.
The bone morphogenetic protein 10 (BMP10) gene spans ~7.8 kb on human chromosome 2p13.3. It is highly expressed in the trabeculae of the embryonic heart, but it is expressed at low levels in the liver and the lungs. BMP-10 belongs to the bone morphogenetic protein (BMP) family.

Biochem/physiol Actions

Bone morphogenetic protein 10 (BMP10) acts as a potent tumor suppressor in prostate, gastric and breast cancer. It is implicated in trabeculation of the embryonic heart. BMP10 retards the growth of prostate cancer cells via, apoptosis induced by X- inhibitor of apoptosis protein (XIAP) and extracellular signal–regulated kinase (ERK)1/2 dependent signaling and SMAD-independent signaling. In addition, it also has an ability to inhibit prostate cancer cell migration and invasiveness. Elevated expression of the gene inhibits growth, adhesion and migration of bladder cancer cells in vitro. Thus, this protein can be considered as a potent therapeutic target for bladder cancer.

Preparation Note

Centrifuge the vial prior to opening. Avoid freeze-thaw cycles.
Reconstitute to a concentration of 0.1-1.0 mg/mL in water containing BSA (50 mg BSA per 1 mg of protein). This solution can then be diluted into other aqueous buffers
