General description

MDC1是粘连蛋白复合物的亚基,它是有丝分裂和减数分裂中姐妹染色单体粘连(sister chromatid cohesion)所需的关键蛋白。该蛋白的温度敏感性突变与姐妹染色单体粘连、染色体凝集和染色体分离的缺陷有关。 其表达受细胞周期调节,在S期达到最高。 MDC1的表达是响应DNA损伤而出现的S- 和 G2/M检查点介导的细胞周期阻滞所必需的。 研究显示MDC1可与DNA双链断裂位点附近的H2A.X 相互作用,并将ATM激酶募集至DNA的损伤区。与这种作用一致的是,MDC1 似乎调节其它涉及DNA损伤检查点的蛋白(如BRCA1和 Chk2)的功能。
