The antibody detected acetylated histone H4 in acid-extracted proteins from HeLa cells. Cross-reacts with acetylated histone H2B from Tetrahymena and weakly cross-reacts with histone H2B from HeLa cells. May cross-react with other acetylated proteins. Using sodium butyrate, an inhibitor of histone deacetylases, enhances detection of acetylated histone H4. For immunocytochemistry, human A431 cells were treated with 5 mM sodium butyrate, an inhibitor of histone deacetylases, for 24 h and fixed with ice-cold 1% paraformaldehyde in PBS. Variables associated with assay conditions will dictate the optimal dilution.
