General description

一种氨基核苷类抗生素,可作为原核和真核蛋白质合成抑制剂。类似于氨酰tRNA的氨酰腺苷酸末端,竞争与大核糖体亚单位的“A位点”结合。其掺入生长中的多肽,导致链延伸终止和新生多肽基嘌呤霉素的释放。用于选择表达转染pac基因(白链霉菌)的哺乳动物细胞系,其产物嘌呤霉素乙酰转移酶通过乙酰化使药物失活。诱导胸腺细胞和HL-60白血病细胞内DNA断裂。转染哺乳动物细胞的推荐选择范围为1至30 µg/ml。
