Anti-Calcium Channel Antibody, Voltage Gated Pan α 1, CT

Purified peptide from {C-DNFDYLTRDWSILGPHHLD [CP15], cooresponding to residues 1506-1524 of rat alpha-1C {Accession P2202} subunit of skeletal muscle voltage-gated calcium channel (Striessnig et al. 1990), conjugated to KLH (Harlow & Lane 1988).

Anti-Calcium Channel Antibody, Voltage Gated Pan α 1, C-terminus detects level of Calcium Channel & has been published & validated for use in IC, IP & WB.

In preparing full-length protein samples for Western blot or immunoprecipitation, the procedures should be performed at 4°C, with the following protease inhibitor mixture: pepstatin A (1 μg/mL), leupeptin (1 μg/mL), aprotinin (1 μg/mL), Pefabloc SC (0.2 mM), benzamidine (0.1 mg/mL), and calpain inhibitors I and II (8 μg/mL each) (Westenbroek et al. 1992).

Western blot: 1:200 using ECL on rat brain membranes.

Immunohistochemistry using rat heart sections. Note: the epitope can be masked by other subunits in some Ca2+ channels.

Dilutions should be made using a carrier protein such as BSA (1-3%)

Optimal working dilutions must be determined by the end user.

Recognizes all types of alpha1 subunits of voltage-gated calcium channel. Does not cross react with any other calcium channel antigens tested so far. The epitope recongnized by the AB5150 is masked by alpha2 and delta chains, so this antibody can bind only after using Triton X 100 or other detergents that cause subunit dissociation (Ahlijanian et al. 1991).
