General description
Activin receptor type-1 (ACVR1) is a member of the protein kinase superfamily, and more specifically to the TKL Ser/Thr protein kinase family and TGFB receptor subfamily. Activins are growth and differentiation factors that belong to the transforming growth factor-beta (TGF-beta) superfamily. ACVR1 plays a role in ligand binding and forms a receptor complex consisting of two type II and two type I transmembrane serine/threonine kinases. The process of forming this receptor complex involves the phosphorylation of Type II receptors; the activation of Type I receptors; the autophosphorylation of Type I receptors, which then bind and activate SMAD transcriptional regulators. ACVR1 is expressed in normal parenchymal cells, endothelial cells, fibroblasts and tumor-derived epithelial cells. Furthermore, ACVR1 is shown to interact with FKBP1A and FCHO1. Defects in ACVR1 are a cause of fibrodysplasia ossificans progressiva (FOP) - a rare autosomal dominant disorder of skeletal malformations and progressive extraskeletal ossification. Additionally, ACVR1 may play a role in the left-right pattern formation during embryogenesis.
