General description
Transcription factor A, mitochondrial (UniProt: Q00059; also known as MtTFA, Mitochondrial transcription factor 1, mtTF1, Transcription factor 6, TCF-6, Transcription factor 6-like 2, TFAM) is encoded by the TFAM (also known as TCF6, TCF6L2) gene (Gene ID: 7019) in human. TFAM is essential for human mtDNA transcription and acts as a key regulator of mtDNA copy number. It is synthesized as a precursor form with a mitochondrial targeting peptide that is cleaved following import into mitochondria. TFAM consists of an amino-terminal HMG domain, a basic linker region, a second HMG domain and a basic carboxy-terminal tail. TFAM binds to the mitochondrial light strand promoter and functions in mitochondrial transcription regulation. It is required for accurate and efficient promoter recognition by the mitochondrial RNA polymerase. TFAM is reported to promote transcription initiation from the HSP1 and the light strand promoter by binding immediately upstream of transcriptional start sites. It is also able to unwind DNA and bend the mitochondrial light strand promoter DNA into a U-turn shape via its HMG boxes. Tissue-specific depletion of TFAM protein is shown to deplete mitochondrial tDNA and causes severe respiratory chain deficiency. (Ref.: Ekstrand, MI et al. (2004). Hum. Mol. Genet. 13(9): 935-944).
