Anti-GOLPH3 Antibody

Golgi phosphoprotein 3 (UniProt Q9H4A6; also known as Coat protein GPP34, MIDAS, Mitochondrial DNA absence factor) is encoded by the GOLPH3 (also known as GPP34) gene (Gene ID 64083) in human. GOLPH3 is a phosphatidylinositol 4-phosphate- (PI4P-) binding protein that plays an essential role in vesicle trafficking and Golgi structure. GOLPH3 is localized to the trans-Golgi via tight binding to PI4P that is highly enriched at the trans-Golgi. GOLPH3 interacts and recruits the unconventional myosin MYO18A to the Golgi, allowing MYO18A and F-actin interaction. The complex applies a tensile force that pulls and deforms the Golgi membrane to facilitate vesicle budding and subsequent vesicles trafficking from the Golgi to the plasma membrane. GOLPH3/MYO18A pathway is reported to mediate DNA-PK-dependent DNA damage response that is essential for normal cell survival and, when enhanced, can confer resistance to killing by DNA damaging agents. GOLPH3 is found overexpressed in many solid tumors and cancer cell lines. Overexpression of GOLPH3 occurs frequently in rhabdomyosarcoma and GOLPH3 knockdown impairs the proliferation of rhabdomyosarcoma cell lines.

Immunocytochemistry Analysis: A 1:250 dilution from a representative lot detected GOLPH3 in HeLa cells.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected GOLPH3 in 10 µg of lysate from untransfected, but not GOLPH siRNA-transfected, HeLa cells (Courtesy of Matthew Buschman, Ph.D. and Seth Field, M.D., Ph.D., University of California, San Diego, USA).

This polyclonal antibody stains Golgi apparatus by indirect fluorescence immunocytochemistry. This polyclonal antibody specifically detected the target band in lysate from control, but not GOLPH siRNA-transfected HeLa cells.
