General description
Eukaryotic cells rely on actin and microtubule-based protein "motors" to generate intracellular movements. These motor proteins contain specialized domains that hydrolyze ATP to produce force and movement along a cytoskeletal polymer (microtubules in the case of the kinesin family and dyneins). In all cell types, dynein is a large protein complex composed of six distinct subunits and is responsible for most intracellular movement toward the minus ends of microtubules. Cytoplasmic dynein has many important roles in transporting membrane bound organelles, including endosomes, lysosomes, and mitochondria. It is also involved in the positioning of the centrosome, the nucleus, and the Golgi. Cytoplasmic dynein is also responsible for virus transport to the nucleus, retrograde axonal transport, and microtubule and neurofilament transport. Three families of dynein light chains (DYNLT) have been identified. Two isoforms of the DYNLT family of light chains, DYNLT1 and DYNLT3, have been proposed to link dynein to specific cargoes. Recent publications indicate that DYNLT1 binds to Rhodopsin and Doc2 whereas DYNLT3 does not.
