一般描述

The protein named Mitochondrial dicarboxylate carrier or Solute carrier family 25 member 10, or Dicarboxylate Carrier Slc25a10 and encoded by the gene named SLC25A10 or DIC is a mitochondrial transport protein that mediates the transfer of malonate, malate, and succinate in exchange for phosphate, sulfate, sulfite or thiosulfate across the mitochondrial inner membrane. These components are critical for the function of the Krebs cycle, as well as gluconeogenesis, urea and sulfur metabolism. Mitochondrial dicarboxylate carrier is expressed in high amounts in the liver and kidney and at lower levels in all other tissues and is localized to the mitochondria and specifically to the inner mitochondrial membrane. Mitochondrial dicarboxylate carrier is also upregulated in various tissues in response to free fatty acids particularly in adipose tissues, and plays a role in mitochondrial malate transport and in the regulation of glucose-stimulated insulin secretion from pancreatic beta cells. Impairment of Mitochondrial dicarboxylate carrier is associated with ethylmalonic encephalopathy (EE) and short chain acyl-CoA dehydrogenase deficiency (SCADD) syndromes. Research indicates that it is the buildup of ethylmalonic acid in the tissues that impairs the Succinate and malate uptake by Mitochondrial dicarboxylate carrier leading to metabolic distress and dysfunction.

Ferroptosis is a programmed iron-dependent cell death associated with peroxidation of lipids particularly, phospholipids. Several studies suggested a possible contribution of mitochondria to ferroptosis although the mechanisms underlying mitochondria-mediated ferroptotic pathways remain elusive. Reduced glutathione (GSH) is a central player

"Redox reactions are powerful chemical processes that involve the reduction and oxidation of proteins and metabolites found in living things. The mechanisms that regulate them are key to maintaining homeostasis and the balance between good health and disease pathology. Oxidative stress is the state where the delicate balance of redox biology is upset, and the pathology of oxidative stress are the cellular consequences to such an imbalance."

"Apoptosis & Autophagy in Cancer: Cell Survival by Eluding Cell Death The ability of tumor cells to elude programmed cell death, also known as apoptosis, is a hallmark of most types of cancer. Similary, autophagy is a highly regulated, homeostatic degradative process where cells destroy their own components via the lysosomal machinery and recycle them for prolonged cell survival. Via extensive crosstalk with pro-apoptotic signaling pathways, autophagy can also contribute to cell death and greatly influence general cell health. Elucidating the correlation between autophagy and apoptotic cell death has become the focus of a great deal of research, particularly in tumor biology. On one hand, autophagy may induce cell death by degrading essential components; on the other hand, it may facilitate survival of cancer cells under unfavorable metabolic conditions."

"Aging: getting older, exhibiting the signs of age, the decline in the physical (and mental) well-being over time, leading to death. Since the beginning of time, man has been obsessed with trying to slow down, stop, or even reverse the signs of aging. Many have gone as far as experimenting with nutritional regimens, eccentric exercises, fantastic rituals, and naturally occurring or synthetic wonder-elements to evade the signs of normal aging. Biologically speaking, what is aging? And what does the latest research tell us about the possibility of discovering the elusive “fountain of youth”? Many advances in our understanding of aging have come from systematic scientific research, and perhaps it holds the key to immortality. Scientifically, aging can be defined as a systems-wide decline in organismal function that occurs over time. This decline occurs as a result of numerous events in the organism, and these events can be classified into nine “hallmarks” of aging, as proposed by López-Otin et al. (2013). Several of the pathologies associated with aging are a direct result of these events going to extremes and may also involve aberrant activation of proliferation signals or hyperactivity. The hallmarks of aging have been defined based on their fulfillment of specific aging related criteria, such as manifestation during normal aging, acceleration of aging if experimentally induced or aggravated, and retardation of aging if prevented or blocked, resulting in increased lifespan. The nine hallmarks of aging are genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, deregulated nutrient sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, and altered intercellular communication. The biological processes underlying aging are complex. By understanding the hallmarks in greater detail, we can get closer to developing intervention strategies that can make the aging process less of a decline, and more of a recline."
