Clone DATK-32 is a rat monoclonal antibody that detects 4β7 integrin/LPAM-1. It targets an epitope from the extracellular domain that is specific to the 4β7 heterodimer.

Preparation Note
0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Integrin beta-7/Integrin alpha-4 (UniProt: P26011/Q00651; also known as Integrin beta-P, M290 IEL antigen/Integrin alpha-4, CD49 antigen-like family member D, Integrin alpha-IV, Lymphocyte Peyer patch adhesion molecules subunit alpha (LPAM subunit alpha), VLA-4 subunit alpha, CD49d) is encoded by the Itgb7/Itga4 gene (Gene ID: 16421/16401) in murine species. Integrins are heterodimeric transmembrane proteins composed of an alpha ( ) and beta (β) subunit that mediate cell adhesion and signaling. They form a large family of receptors with 24 unique β combinations, each demonstrating specificity for distinct ligands and are critical for a wide range of biological processes. Integrin β heterodimers are divided into four classes based on evolutionary associations (leukocyte, collagen-binding, Arg-Gly-Asp (RGD)-binding, and laminin-binding integrins). 4β7 integrin/LPAM-1 mediates lymphocyte adhesion to high endothelial venules (HEVs) in Peyerβs patches (PPs) and lamina propria, facilitating immune cell recruitment to mucosal sites. It selectively regulates lymphocyte trafficking and adhesion in the gut and gut-associated lymphoid tissue (GALT). 4β7 plays a specialized role in intestinal immunity, enabling B cells to home to GALT, where they undergo class switching and differentiation into immunoglobulin A (IgA)-secreting plasmablasts. 4β7 plays a pivotal role in lymphocyte adhesion to various ligands, such as MAdCAM-1, VCAM-1, and fibronectin, and supports aggregation of lymphocytes during immune responses. Reduced 4β7 expression or impaired CD22-Shp1 signaling disrupts intestinal immunity, leading to diminished antibody responses and pathogen resistance.
