General description
单克隆抗GAPDH(小鼠IgM同种型)源自通过小鼠骨髓瘤细胞(NS1细胞)融合产生的杂交瘤GAPDH-71.1和通过兔GAPDH免疫的BALB/c小鼠脾细胞。
甘油醛-3-磷酸脱氢酶(GAPDH)的基因定位于人染色体12p13。该蛋白定位于细胞质中,但可根据细胞条件转移至细胞核。

Biochem/physiol Actions
甘油醛-3-磷酸脱氢酶(GAPDH)是含有相同链的四聚体。它可催化甘油醛-磷酸盐的可逆氧化磷酸化,其碳水化合物代谢中的关键能量产生步骤。它可与几种蛋白质结合,包括肌动蛋白、微管蛋白、淀粉样蛋白前体、聚谷氨酰胺肽、DRPLA(齿状核红核苍白球丘脑下部核萎缩)和亨廷顿蛋白。蛋白激酶Cι/λ可结合并磷酸化GAPDH。磷酸化的GAPDH可与细胞骨架元件结合并控制早期分泌途径中的微管动力学。聚(ADP-核糖)聚合酶-1(PARP1)可与GAPDH相互作用,从而在氧化/亚硝化应激的存在下介导脑损伤。GAPDH形成OCA-S的一部分,其中OCA-S是多组分OCT1(八聚体-基序-结合因子)共激活因子复合物,其可参与S期依赖性组蛋白H2B的转录。该关联可将H2B转录机制与细胞周期调节和细胞代谢状态联系起来。GAPDH也是功能性GAIT(干扰素γ激活的翻译抑制剂)mRNP(信使核糖核蛋白)的组分。在黑素瘤进展期间GAPDH表达失调。
