General description
The gene NFAM1 (NFAT activation molecule 1) is mapped to human chromosome 22q13. It is mainly expressed in spleen B and T cells. The encoded protein has a signal peptide, a transmembrane region and an immunoglobulin domain in the extracellular region.

Biochem/physiol Actions
NFAM1 (NFAT activation molecule 1) controls B cell signaling and development via ITAM (immunoreceptor tyrosine-based activation motif). It participates in the activation of the calcineurin/NFAT (nuclear factor of activated T-cells)-signaling pathway and thus controls downstream cytokine gene promoters. In Paget′s disease of bone, NFAM1 is associated with osteoclast formation and bone resorption activity. It controls intracellular calcium levels, PLCγ (phospholipase C), expression of calcineurin and activation of Syk (tyrosine kinase).
