General description
Ras and Rab interactor 3 (RIN3) is a novel Rab5-binding protein, encoded by the gene mapped to human chromosome 14q32. The encoded protein belongs to the RIN family of RAS effectors. RIN3 contains a SH2 domain, proline-rich domain, RH domain, vacuolar protein sorting-associated protein (Vps9) domain and ras-associating (RA) domain in the N- terminal region. RIN3 show tissue-specific expression and is highly expressed in mast cells.

Biochem/physiol Actions
Ras and Rab interactor 3 (RIN3) plays an essential role in vesicular trafficking by interacting with small GTPases such as Ras and Rab14. In addition, it also interacts with amphiphysin II and promotes receptor-mediated endocytosis. Mutations in the gene increases the risk of susceptibility to Paget′s disease of bone (PDB). The encoded protein acts as a guanine nucleotide exchange factor (GEF) for Rab 5 and Rab31. RIN3 plays an important role in basic mast cell functions and pathologies such as mast cell mediated chronic inflammation and mast cell hyperproliferation by regulating receptor tyrosine kinase (KIT) activity.
