General description

抗黏着斑蛋白单克隆抗体（小鼠 IgG1 亚型）来自于免疫BALB/c小鼠骨髓瘤细胞和脾细胞相互融合而产生的hVIN-1杂交瘤细胞。黏着斑蛋白位于闰盘（心肌）的粘着膜、肌腱连接（骨骼肌）、神经肌肉接合部位以及膜相关的平滑肌致密体中。在经过病毒转化的许多细胞类型中，黏着斑蛋白被重新分配至玫瑰花结或伪足小体。

Biochem/physiol Actions

使用黏着斑蛋白单克隆抗体，通过间接免疫荧光标记法，能够对组织和培养细胞物中的细胞间和细胞-基质连接处的黏着斑蛋白进行特异性染色。在免疫印迹中，抗体与116kDa的黏着斑蛋白条带反应。本品可以与许多物种的黏着斑蛋白反应。与人、牛、鸡、狗、大鼠、小鼠、火鸡和非洲爪蟾具有良好的反应性。该抗体与平滑肌黏着斑蛋白剪切变体metavinculin具有交叉反应性。

特异性标记细胞间和细胞-基质连接处的黏着斑蛋白。与人黏着斑蛋白反应强烈。与平滑肌黏着斑蛋白剪切变体metavinculin具有交叉反应性。

黏着斑蛋白是一种细胞骨架蛋白，与细胞间和细胞-细胞外基质粘附连接的细胞质表面相关。它是将F-肌动蛋白锚定到细胞膜上的几种相互作用蛋白之一。一系列分子相互作用可能参与黏着斑的跨膜组装。在黏着斑的组装中，整合蛋白的β亚基与踝蛋白结合。踝蛋白与黏着斑蛋白结合，黏着斑蛋白与α辅肌动蛋白相互作用并且可能与其自身相互作用。由于α辅肌动蛋白与肌动蛋白微丝结合并交联，因此黏着斑蛋白是细胞外基质与细胞质微丝系统的跨膜连接中的关键元素。
