Extracellular matrix protein 1 (UniProt: Q16610; also known as Secretory component p85) is encoded by the ECM1 gene (Gene ID: 1893) in human. ECM1 is a secreted glycoprotein that is involved in cell proliferation, angiogenesis, and cell differentiation. It is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature protein that is involved in endochondral bone formation and serves as a negative regulator of bone mineralization. It is shown to interact with a variety of extracellular and structural proteins, contributing to the maintenance of skin integrity and homeostasis. Overexpression of ECM1 has also been reported in several cancers, including pancreatic cancer, breast cancer, gastric cancer, and hepatocellular cancer. It leads to poor prognosis in cancer patients. Its over-expression is also observed in colorectal cancers (CRC) where it is correlated with tumor size, lymph node status and Tumor, Node, Metastasis (TNM) stage of cancer. Its knockdown suppresses CRC cell growth, migration, and invasion. The occurrence and progression of CRC by ECM1 reportedly mediated through the PI3K/AKT/GSK3β/Snail signaling pathway. Mutations in ECM1 gene have been linked to lipoid proteinosis that is characterized by generalized thickening of skin, mucosae. and certain viscera. Four isoforms of ECM1 have been described that are produced by alternative splicing. Quality Control Testing

Evaluated by Western Blotting in Human testis tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ECM1 in Human testis tissue lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ECM1 in human epididymis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound ECM1 peptide with a KD of 3.3 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ECM1 in MCF-7 cells.

Clone 1C6 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects Extracellular matrix protein 1 (ECM1). It targets an epitope within 25 amino acids from the N-terminal region.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
